Last week, the University of Guelph published findings of their study on different varieties of melatonin. The university tested 30 different melatonin supplements from 16 brand names. The supplements were sold in grocery stores and drug stores. Of the products tested, 71 percent did not meet their label claims. Some examples of these inaccuracies are listed below:
- 12-25 percent had higher than listed melatonin amounts
- Some products contained almost 5 times the listed amount of melatonin
- One chewable tablet that listed 1.5 mg of melatonin actually contained over 8 mg of melatonin
- Some products skewed the other way, containing less melatonin than listed
- One product listed 3 mg melatonin but contained 83 percent less
- One quarter of the supplements also contained serotonin, which is not authorized to be sold as a supplement, as it could be dangerous for those on SSRI’s.
In the article, doctors commenting on the findings noted that as an unstable molecule, melatonin could be affected by environmental conditions (such as temperature) or differences among batches (such as by improper mixing). The doctors speculated that feasibly some manufacturers may have added extra melatonin in an attempt to extend the product’s shelf life. Their final conclusion was their hope that these findings could help manufacturers improve production quality.
Sadly, findings such as these are not new, and confirm yet again what we have said all along. Raw material quality matters and manufacturing should be performed responsibly. Life Choice only uses USP pharmaceutical grade raw materials and manufactures at GMP facilities.
There are 3 grades for raw material: pharmaceutical (which meets pharmaceutical standards of purity 97.5%+); food grade, which meets the standards for human consumption (more added fillers); and feed grade, which meets the standards for animal consumption (low quality, not much regulation). Another reason for the huge difference is the large batch blending and the improper distribution of the active material. If we consider the active as being 3 mg, when encapsulated in the standard size “2” capsule, the total fill weight will be 370 mg with 367 mgs of fillers, and in tablets, the fillers are even more. If encapsulated in size “0,” the total fill weight would be 680 mg or 677 mg of filler. In perspective, a large pharmaceutical company would run a minimum of 100 million plus capsules, using 300 kgs of melatonin and 36,900 kgs of filler. Some companies use Crospovidone (cross linked polyvinyl N-pyrrolidone, or PVP) in their melatonin. PVP is a common inactive ingredient used by the pharmaceutical industry. While its swelling properties make it useful for absorbing active drugs, PVP is not absorbed orally. Pulmonary emboli have been reported in autopsies of intravenous drug abusers who have crushed tablets, such as hydromorphone, for injection. The long-term effects of crospovidone in the lung are unknown, but topical application has resulted in allergic reactions, also.
It is perfectly understandable why each capsule is vastly different in the melatonin content, for blending equally is next to impossible. This lower yield gives an equal distribution of melatonin per capsule.
